Choose a handgun based on the needs. I personally want light recoil in order to get the sights back on a threat faster, very basic control layout with no exposed hammers and safeties, as much ammo storage without comprimising its concealibility, along with a light attached for an additional element I may possibly blind the threat with.
An important element to understand, that 9 out of 10 times any weapon is presented, is generally enought to de-escalate a threatening encouter.
A defensive firearm will only be effective if you carry upon your person in a time of need. I would have no problem sacrificing size and cal in order to have a small enough handgun I would be sure to always have with me.
The smaller the bullet size, the more dependant will be its effectiveness upon accuracy. Practice, practice...and practice.
Practice real life carrying, and then engaging a threat from close up. Mimic an encounter by presenting , and then moving away from a threat while engaging.. Hopefully off line from them, not just straight back.
The above is just my opinion, and I encourage the above techniques to only be done safely, and with dry parctice first.
